It's possible, especially if you use a mixer upstream of the conversion to USB, but, in general, you cannot plug two microphones into the game directly and have two people sign up as singers in QuickPlay or Tour mode.

However, I do believe that Duel mode uses two microphones.

Duel mode does support two mics (plus controllers).

Some have used dual mics from Karaoke Revolution - two 6.5mm jacked mics that plug into a single USB box. You can pick these up off of eBay for less than $20. There is still only one vocal track and on-stage vocalist in-game, but it allows two players to sing (thus opening up your party gaming to include five people at once).
