I have two headsets for the 360 and one mic.

So if I want to play with vocal harmonies, can I plug in one headset in my guitar and the second headset in my second guitar for my friend?

no, you cannot. You can, however, plug the mic in the 360, and plug the headset into the controller that controls the vocals.

You can't use headsets to sing in TB:RB.
