I'm guessing it's not a PS3 disc then... those Blu-Ray discs are easy to clean and pretty damn tough.
If it's a standard DVD and has some scuffs or scratches on it, there are places that can "resurface" the disc. Your local video store should have one of those machines that can resurface/buff discs.

If there's no deep marks or scratches and you think it's just dust, I find the best way to clean a disc (BD/CD/DVD) is to grab yourself a lens cloth (like what you clean specs/glasses or LCD screens with), hold the disc close to your mouth and as you use your breath to steam up a section of the disc, wipe it with the cloth (in a straight line from the inner ring to the outside), keep spinning the disc around, steaming it up and wiping it off, and eventually you'll get all the dust off.
If it's got gunk stuck to it (i've seen my share of rental discs and wonder what the hell people do with them... fingerprints, food, you name it)... you can rinse the disc under cold water and while you're doing that, gently rub off anything that may be stuck to the surface of the disc. Let it dry on its own and then use the lens cloth technique above to clean off any watermarks and dust.
Some say to use toothpaste, metal polish, sticking it in boiling water or in the freezer, but I wouldn't recommend any of those methods.
If the disc isn't scratched or marked and still doesn't work, the only way to save it is to get it buffed/resurfaced. Most video rental and some music stores will probably have a machine that does that, and they often only charge a couple of dollars to do it. Once the disc's resurfaced, it should be as good as new.
