...I have noticed that an odd pain appears with my fret hand after trying to speed all around the buttons on fast parts....I know I could try using the solo buttons, but my hands are kinda big so its very awkward.

Any of you have this pain?

Without knowing more details about your pain, I'll just say that if you don't play real guitar regularly or haven't played the Guitar Hero games on expert for some prolonged period of time, your wrist/hand are going to be sore from playing Rock Band on expert. Just take a break after a while... maybe ice your wrist/hand if you think it's necessary. Like any other new hobby/sport, it takes time to build up stamina. I'm not a drummer, and my calf is sore today from Rock Band Drums. Just don't overdo it.

Silly as it seems, I think most advice for playing painfree on a real guitar will probably help you play your plastic one. The first tip that comes to mind is that your palm should not be touching the neck of the guitar. Instead your wrist should (ideally) be bent slightly towards you and the body of the guitar.

You could try googling for guitar technique and see if you can glean anything that would be useful for your Rock Band playing.

Keeping your thumb on the "Center" of the back of the neck will go a LONG away to help you in preventing alot of wrist strain.
And if your arm is getting tired on the strumming, then I would suggest learning to Alt Strum using your wrist instead of your arm doing all down strumming.
The same rules apply for a real guitar as well.

I'm a Bass player IRL, so I know where you're coming from with the upper fret buttons being too close together (fat finger syndrome). Takes practice just like everything else I guess.

The pain or discomfort rather, isn't in the wrist really....its right around the area of my index and middle finger....around the knuckles. Feels like they are trying to pop or something. Very strange.

It's never happened with GH or anything, just expert on this game.

Maybe I am not used to "that" guitar in particular.

Thanks for the advice though guys, its always appreciated. :)
