I really hope that when you're doing vocals there's some sorta quick test they can take and it'll tell them to speak up or quiet down. Because while that may be cool for single player or in a party atmosphere, I get the feeling that it could really start to annoy if you're doing online multiplayer, and you're constantly getting lead singers whose volume levels are out of whack with all the other instruments.
Just adjusting the levels in-game probably won't cut the mustard since that won't account for people distorting the mic or having everything sound like a whisper (although perhaps the scoring function based on the tone they hit will force them to do it right, I dunno?)
Forgive me if this kinda thing is already a given with karaoke games, but I've never played one before and just started thinking

That's an interesting thought, I hadn't really considered that. Maybe there's a way to digitally equalize the voice with the other instruments.

Probably the best way to do it is simply to have a preference set thing, where they have a vocal track, and you use it to calibrate the highest and lowest points you want vocals to be, and the system automatically adjusts to keep it in the range

When you get rid of all the peaks in volume in a signal it's called compression. Basically all of the loud sounds are compressed down to a lower level closer in volume to the quiet stuff. Then the compressed sound is turned up so that you can hear everything equally.
This is how a singer can sing or whisper one verse and then scream the chorus and it all sounds the same volume on the album (IE the Deftones). I'm sure that this will be built into the game hardware.

They could do it really quick at the beginning of a song. Call it a Sound Check.
