Yes, but I'm not sure if its restricted in combination. I've seen IGN do guitar and mic.

Yes, no restrictions but common sense. If you can play drums and guitar at the same time because you're a mutant with 4 arms, more power to you :) It's treated as 2 separate players in a normal band; there's no special "guitarist/vocalist" mode.

I actually did this last night at Best Buy. It's tricky and you need to know the words to the songs (or just hum when you forget...) but it's totally possible. You will be rendered as two characters on screen, though.
