One overlooked instrumental is Into the Lungs of Hell by Megadeth. Give it a listen, it's really underrated and (IMO) better than some of Metallica's instrumentals.

Ignoring background vocals
The Calling by Santana and Clapton.
And some others (some of which have been mentioned)
White Cliffs of Dover by Eric Johnson
For the Love of God by Vai
Summer Song by Satriani (and like Vai could have many others)
Eruption - Van Halen. Yeah I know it's just an extended solo, but I like it.

Some good ones have already been mentioned.
Schiz mentioned Ventures. Other great ones from that era:
Dick Dale. Misirlou
http://www.youtube.com/watch?v=ZIU0RMV_II8
First speed metal song?
Duane Eddy. Probably my favorite of his is Rebel Rouser
http://www.youtube.com/watch?v=Br5B2ZaO79E
Link Wray, Rumble
http://www.youtube.com/watch?v=PP0aRKnyzpc
Jeff Beck has two of my favorites -- Beck's Bolero (disputed between Page and Beck who wrote it, seems pretty clear both had a hand to my ear. The main melody is much more Jeff's style, but the opening chords are definitely Jimmy's and the riff in the break sounds like him, too. No question Jimmy was the better and more prolific writer, though. But I prefer Jeff's playing.)
http://www.youtube.com/watch?v=Xbx-Qgt65bI
And 'Cause We've Ended As Lovers, which I don't think I need to post.
Clapton's version of Freddie King's Hideaway from his recording with Mayall's Bluesbreakers is a gold standard of blues instrumentals.
Eric Johnson's Zap is the song that was his breakthrough
http://www.youtube.com/watch?v=W0qBH-SWXVY
I like the Satriani and Vai selections that have been mentioned.
I love Steely Dan's cover of Duke Ellington's East St Louis Toodle-Oo. Awesome pedal steel by Skunk Baxter.
http://www.youtube.com/watch?v=br4mB7VWf7E
Here's a combo that'll stand up to any comparison: Larry Carlton and Robben Ford

Santana is amazing, I was listening to my Abraxas 8 track the other day....
How about Santana's (i believe) 2 instrumentals on Supernatural? I liked them...
and Chester and Lester are both amazing guitarists (I posted a vid of them playing together in the overated guitarists thread (not as overrated but to shut someone up about shredders being the best))
as for flamenco, im only starting to get into that due to Paco de Lucia, but everything ive heard by him is fantasic, prolly the fastest guitarist ive ever heard
as for more instrumentals that are guitar driven, how about some Roy Rogers?

If you want to go that route, then Koyunbaba is the way to go. It's this sweet classical piece I heard when I went to a classical show. It's an old, OLD, jam, but oh so sweet. If you can find it, you're in for a treat.

As far as instrumentals go then, I love YYZ. I know everyone hates the synth intro, but I don't care, I can't picture the song without it. It has an awesome guitar solo that is extremely hard to play (I can only play about half of it).
Moby Dick by Led Zeppelin has awesome riffs in it too, but it primarily features Bonham's drums so does that count? Does in my book.

Ah jeez -- you guys jammed? :( D@mn these separate platforms.
-- I appreciate what SRV did, but Jimi's is like a Monet. A perfect little gem.
My favorite version though is the Gil Evans big band arrangement. Really emphasizes the beauty of the song, not so much the awesome guitar playing.

I'd have to go with Herbie hancock's Watermelon Man and Cantaloupe Island -actually, the whole Headhunters, Fat Albert Rotunda and Empyrean Isles albums would be more precise.
Also, Death In Vegas "Ein Dur Die Damen"...
Which brings me to kraut, so I cannot miss Neu! "Hallogallo",
And, finally, even though there are some very minimal lyrics, Kraftwerk's "Autobahn". Since the whole lyrics are "Wir fahren auf der Autobahn" and the song is 23 minutes long, it can be considered an instrumental.
edit : okay, "guitar-driven", says the topic. Nevermind, then, not one of these songs has guitar
