So I've been listening to Give Me Shelter, and I'm wondering how Harmonix is gonna deal with the woman singing, because her voice cracks pretty severely during part of that song. It seems like a minor question, but I'm just curious.
It's also gonna be ackward to sing "Rape, Murder" three times in front of my friends and/or family.

You think that's awkward, I just sang it in front of a crowd at Times Square yesterday! Multiple times!
I think the note tubes are set to have you singing without the cracking. I didn't really notice when I was singing it.

Someone who's played the retail version can answer this better than I can, but I'll give it a shot.
Guess 1: Backup vocals are for backup singers. The Rock Band singer will be taking lead and sing Mick's part only.
Guess 2: The original album version of the song didn't have the background singers so HMX may have gone that route.

And you're 0 for 2, sorry. You do have sing the backup singers' parts, and the version here has the backup singers.

The original recording of this song on "Let it Bleed" DID have Merry Clayton singing the back up vocals. In addition to that, she also sings the "rape, murder..." solo three times where Mick does not sing at all.
Also, the song is "Gimme Shelter," not "Give Me Shelter."
And as a funny tidbit, Keith Richards actually sang this song on the original recording but the band didn't like how it sounded so they re-recorded it with Mick singing. I have the Keith version and it is pretty interesting.

Damn me and my proper grammar.
