I havnt tried this myself, as I have tried it with a GH controller and there was too much resistance to make it work, but Im thinking the RB controller could easily be used with a guitar pick with the way it was designed. Im even wondering if that was atleast in some way, an intended use. So if anyone has tried it yet, or has thought of it let me know, Ill try when I get home later.

I've messed around playing it with my fingers shaped like I was holding a pick, hitting it with my pointer finger for down-picks and hitting it with my thumb for up-picks, but it was still really awkward to play. I haven't actually tried it with a pick, yet, but that'd probably take just as much getting used to, if not more. It would almost definitely work, but I wouldn't personally do it.

I vaguely remember reading somewhere that the strum bars shape was changed slightly (more bulbous at the end I believe) to make it more comfortable for players who prefer to hold the strum bar when playing. I don't think any of these games were designed for use with a pick really.

I'm going to try it out, GHIII wouldn't allow me to play in the way I did for all the others in the series (thumb and forefinger together, striking down with nail on index finger) and it's ruined me, I can't stand to play like that anymore and have to use my thumb. A pick would be an improvement.
GHIII was horrible preparation for Rock Band...RB's solos are killing me since I've gotten used to GHIII's imprecise "just move your left hand as fast as you can" timing.

I recall HMXSean responding to a post similar to this. The person wanted to know if playing with a pick was an option. Sean had said that it was play tested with a pick and they didn't paint the strum bar to avoid a piick scratching it.
I tried searching for the post and couldn't find it though.

Been a guitarist most of my life and I actually play with my finger and index finger in the shape of a pick. I can strum alternately (up/down) with very little pressure on the strum bar. This seems to work best for me. When I actually play my real guitars, I tend to dig in heavily as I have a very agressive style of playing. This would probably end up killing the strum bar I am certain.
