Yes, but:
1. It has to be plugged into a standard controller, not your guitar controller, and
2. It won't be very good. I tried doing exactly that after getting home from Guitar Center and discovering that the mic stand clip I had bought was way too small for the Rock Band mic. I usually play with the original singer off and my singing cranked up to maximum volume, so I could hear what the game was registering. It was very tinny, very off, and while I managed to barely make it through the Battle of the Vans (3 stars on Blitzkrieg Bop with 2 singer failouts, 4 stars on I Think I'm Paranoid, both vocals/medium and guitar/expert), it was such a struggle against the hardware that I stopped trying afterwards and decided to wait until I could get a proper mic clip. Note that it's not that I'm a bad singer/player--I pulled a solid 5 star on In Bloom with drums/hard and vocals/medium with someone else holding a proper mic.

Or try a reg controller and the wireless headset if you have one....though it cut's out on high notes sometimes....But if you tweak the mic sensitivity it can work out.
