First night I play Rock Band, I do world tour and the guitar is missing all the notes, drum seems to be working fine.
Next night I play w/o the drums and the guitar works perfectly.
Is this a calibration thing?

It could be that you were simply not used to the Strat, and playing early or late. It may also be because of your drummer being slightly early or late.

Sometimes I find that happening in songs on Gh3 too, sometimes your sight/action delay may be off due to sleepiness. Get some sleep and play again, if you keep on having the problem send your guitar back in.

i appreciate the feedback, but unfortunately it's definitely not the issue.
i've beaten most of the songs on hard in the solo tour with the guitar, but when i play medium, i'm failing a minute in, though i'm absolutely hitting the notes.
guess that's why i asked if it might be a calibration thing since it works one way but not the other.

I'm having the same problem. When I play with the drums, the guitar lags so bad I struggle to finish a song in Medium.

try messing with calibration. My tv is a rear projection and i tried self calibrating and everything and nothing seemed to work. LCD was the right choice for some odd reason. My problem was drums worked perfect and guitar was off. The new patch needs to have it so each instrument can have different calibration. Even though that shouldnt be the case, it is

I just jumped on this forum to see if others were having the same issue. It's almost like the drums are interfering with the guitar side. If someone is playing drums, the guitar will skip notes, drop out of long notes and do all sorts of craziness. Definitely an issue.

Are they both plugged into the USB hub? If so, are all the cords connected firmly?

Nope, both are plugged into individual USB slots on the front of the 360.
