When RB3 came out, I actually thought maybe they had built the Road Challenges like that so you could "bolt on" new tours, art assets, and venues via DLC packs. But, the feedback we got at the time seemed to indicate RB3 wasn't built for support of that kind of DLC.
If RB4 happens, the ability to extend the game through DLC Tour Packs is definately up there on my list of wants.
You know, I thought the same thing, especially after having playing thru all the Road Challenges by the second or third day of owning the game. But, alas. I still scratch my head a little over that because it was one area that I felt RB 3 went backward instead of forward on. RB 2 had the ever changing (with new DLC purchases) Tour Challenges. It seems to me that the Road Challenges might have easily incorperated that same idea instead of the stagnant no-real-reason-to-play-it-again version. I also think that would have made them a much better, or at least a more interesting, replacement for the World Tour than it currently is. Hell, it wouldn't even have required Road Challenge DLC if they would have been coded to expand and grow similar to the Tour Challenges.

I get what you're saying here, but I have to disagree with you on the aspect that RB3 had the smoothest gameplay in the series. While yes, it has the most features, there's still some kinks that could be fixed. For example, the current "lane" feature doesn't work the way it's supposed to, and sometimes results in frustrating gameplay.
They could add a better tour mode, maybe even an RPG-style thing where you choose where your band goes, whether you become superstars or underground hits. Maybe they could introduce new game modes and party options, much like how Guitar Hero 5 tried, but failed with the Rockfest multiplayer setups and instant gameplay on the start screen. Maybe Rock Band could feature new ways to play.
And even with Rock Band's extensive soundtrack, we still have some really great, on-disc soundtrack worthy songs we can get. I'd some, but I don't want to take up even more time.
The way they're doing keyboard is a great start. However, restricting it to one hand kinda takes a but of edge out of the experience. Maybe they could come up with a system for two handed charts, though this is unlikely due to backward compatibility issues.
This isn't a great list of things Rock Band 4 could do, but it's a start.

If a RB 4 were to be made, I'd like this as well. I'd actually go as far as making it so you could actually totally fail - or maybe just give yourself a huge setback resulting in a bandmate quitting, or having to play in smaller venues for less money, loosing a promoter, etc.. RB is a game after all, and games need the risk and reward factor to be truly engaging, I think. RB 3 sort of did away with all risks and has nothing but reward. That sounds like a good thing in a way - until you realize that the lack of risk made the rewards feel a little empty.
